Abstract

Purpose – The aim of this study is to determine the differences in the antimicrobial activity of katuk leaf (Sauropus androgynus (L.) Merr) against Escherichia coli.

Design/Methodology/Approach – The method used in this study was experimental posttest using a control group design. Analysis of the effect of katuk leaf was performed in the dilution method with 20%, 40%, 60%, 80%, and 100% concentration. The data were analyzed using one-way ANOVA test (α = 0.05) and was then tested using the least significant difference (LSD) test.

Findings – Bacterial colony counting that used total plant count found the average of E. coli amount at 20% of concentration (526.820 CFU/ml), 40% of concentration (449.380 CFU/ml), concentration of 60% (255.710 CFU/ml), concentration of 80% (194.110 CFU/ml), and at concentration 100% (168.600 CFU/ml). This study concluded that the katuk leaf extract at 20%, 40%, 60%, 80%, and 100% of concentration had antimicroba effect with significant influence. The 100% of concentration had the most significant effect compared with the other concentrations.

Research Limitations/Implications – Katuk leaf could be used as one of the alternative herbal choices that has a compound antimicrobial effect.

Originality/Value – This study increases the theoretical understanding of the difference of antimicrobial effectivity of katuk leaf extract (S. Androgynus (L.) Merr.) concentration against E. coli

Abstract

Purpose

Food and water safety is a major international concern. Among the food groups implicated with greater frequency in recent years as having caused or been associated with enteric diseases in humans are raw fruits and vegetables. Outbreaks of diseases caused by infectious and toxigenic bacteria as well as parasites and viruses have been documented to occur as a result of consumption of contaminated produce. This paper seeks to review the scientific literature reporting evidence to support the potential for preharvest contamination of fruits and vegetables intended to be eaten raw.

Design/methodology/approach

Sources of preharvest contamination of produce include manure, manure compost, sewage sludge, irrigation water, runoff water from livestock operations, and wild and domestic animals. Literature was reviewed to assess the conditions affecting survival of pathogenic microorganisms originating from these sources in preharvest environments and potential for contamination of produce before or at the time of harvest.

Findings

Prevention of preharvest contamination is essential to minimizing the risk of illness caused by consuming raw produce because postharvest treatment with sanitizers cannot be relied upon to eliminate pathogens.

Originality/value

A better understanding of the behaviour of pathogens in preharvest environments will enhance the prospect of developing effective strategies and interventions that will assure the delivery of safe produce to the consumer.

Abstract

Purpose

The purpose of this review is to address the consumer’s preferences that have varied greatly in the past decade appraising the use of flavor and aroma compounds in the development of functional foods rather than consuming artificial additives. A growing interest in natural flavoring agents and preservatives have made the researchers to explore the other bio-functional properties of natural flavors beyond their ability to give a remarkable flavor to the food.

Design/methodology/approach

In this review, five major flavoring agents used significantly in food industries have been discussed for their bioactive profile and promising health benefits. Vanilla, coffee, cardamom, saffron and cinnamon, despite being appreciated as natural flavors, have got impressive health benefits due to functional ingredients, which are being used for the development of nutraceuticals.

Findings

Flavoring and coloring compounds of these products have shown positive results in the prevention of several diseases including carcinoma and neurological diseases such as Alzheimer’s and Parkinson’s. Such effects are attributed to the presence of phenolic compounds, which possesses free radical scavenging, anti-inflammatory antiviral and antimicrobial properties. These properties not only show a preventive mechanism against diseases but also makes the food product shelf-stable by imparting antimicrobial effects.

Originality/value

This paper highlights the opportunities to increase the use of such natural flavoring agents over synthetic aroma compounds to develop novel functional foods. Phenols, carotenoids and flavonoids are the major health-promoting components of these highly valued aroma ingredients.

Abstract

Purpose

The purpose of this paper is to evaluate the protective effects of Persian gum and gum Arabic on the survival of Lactobacillus plantarum subsp. plantarum PTCC 1896, Escherichia coli, Xanthomonas axonopodis, and Saccharomyces cerevisiae during freeze drying.

Design/methodology/approach

Cultures were harvested from the early stationary phase and enumerated after dilution according to the Milse Misra method. Bacterial suspensions were mixed with protective agents and frozen at –80°C before drying in a freeze dryer. Survival rates were determined both immediately during freeze drying and after 14 days of cold storage (at 4°C).

Findings

Compared to gum Arabic 5 and 10 percent (W/V) or skim milk 10 percent (W/V), Persian gum 1 percent (W/V) showed no significantly different effects on the survival of Lactobacillus plantarum subsp. plantarum PTCC 1896 (p<0.05). Similarly, no significant differences (p<0.05) were observed between Persian gum 6 percent (W/V), gum Arabic 6 percent (W/V), the combination of Persian gum 3 percent (W/V) and gum Arabic 3 percent (W/V), and skim milk 10 percent (W/V) in terms of their effects on the survival of Escherichia coli. Skim milk 10 percent (W/V) was, however, found to have significant (p<0.05) effects on the survival of Xanthomonas axonopodis and Saccharomyces cerevisiae. Statistically significant (p<0.05) effects were observed after 14 days of cold storage (4°C) by Persian gum 6 percent (W/V) on the survival of Escherichia coli and by gum Arabic 6 percent (W/V) on the survival of Xanthomonas axonopodis and Saccharomyces cerevisiae. It was concluded that protective agents could be replaced by Persian gum for its effect on the survival rate of Escherichia coli and by skim milk for its effects on the survival of Xanthomonas axonopodis and Saccharomyces cerevisiae. Persian gum 6 percent (W/V) seemed to be the best protective agent for Escherichia coli during 14 days of its storage as gum Arabic 6 percent (W/V) seemed to have the same performance for Xanthomonas axonopodis and Saccharomyces cerevisiae. Persian gum 1 percent (W/V) was also found an alternative protective agent for the freeze drying of Lactobacillus plantarum subsp. plantarum PTCC 1896.

Originality/value

As Iranian Persian gum is cheap due to its wide availability and seems to have effects similar to those of gum Arabic and skim milk at low concentrations, it may be considered a good candidate for industrial applications.

Abstract

Purpose

This study aims to explore the traditional plant dyeing of Xinjiang Atlas silk fabrics, providing references for the comprehensive utilization of plant dyes in intangible cultural heritage.

Design/methodology/approach

The focus of this study is on dyeing experiments of Atlas silk fabrics using safflower extracts, constrained by regional resources. Safflower dry flowers grown in Xinjiang were selected, rinsed with pure water and rubbed. Yellow pigments were removed by adding edible white vinegar. Red pigments from safflower were extracted using an alkaline solution prepared with Populus euphratica ash, a special product of Xinjiang. The extraction rate was analyzed under varying material-to-liquor ratios, pH values, times and temperatures. Direct dyeing process experiments were conducted to obtain different colorimetric L, a, b and K/S values for comparison. Samples with good color development were selected to test the impact of dyeing immersions on color development, and their color fastness, UV protection and antibacterial effects were verified.

Findings

The dyeing experiments on silk fabrics confirmed their UV protection capabilities and antibacterial properties, demonstrating effectiveness against E. coli and Staphylococcus aureus. As a major producer of safflower, Xinjiang underscores the significance of safflower as an essential plant dyes on the Silk Road. This study reveals its market potential and suitability for use in the plant dyeing process of Atlas silk, producing vibrant red and pink colors.

Originality/value

The experiments indicated that after removing yellow pigments, the highest extraction rate of red pigment from safflower was achieved at a pH value of 10–11, a temperature of 30°C and an extraction time of 40 min. The best bright red color effect with strong color fastness was obtained with a material-to-liquor ratio of 1:20, a temperature of 40°C and three immersions. The best light pink color effect with strong color fastness was a material-to-liquor ratio of 1:80, a temperature of 30°C and two immersions.

Abstract

Purpose

Broad-spectrum cephalosporin resistance is rapidly increasing in Escherichia coli, representing a food safety problem. The purpose of this paper is to characterize eight extended-spectrum-ß-lactamase (ESBL) and acquired AmpC ß-lactamase-producing E. coli isolates and virotypes associated, obtained from chicken and pork food samples in Puebla, Mexico.

Design/methodology/approach

Samples (36 from chicken and 10 from pork) were cultured on Levine agar plates supplemented with cefotaxime (2 mg/L) for isolation of cefotaxime-resistant (CTXR) E. coli. CTXR-E. coli isolates were detected in 33 of 46 samples (72 percent), and one isolate/sample was characterized (28 from chicken and 5 from pork), for ESBL production, phylogenetic group, sequence typing, resistance and virulence genes by PCR and sequencing.

Findings

Results showed 16 ESBL-E. coli (35 percent) (12/16 belonging to phylogroup B1) and 8 CMY-2-E. coli (17 percent). ESBL detected were as follows (number of isolates): CTX-M-2 (8); CTX-M-1 (2); CTX-M-15 (1); SHV-2a (4) and TEM-52c (1). In total, 20 different sequence types (STs) were identified among the ESBL- or CMY-2-producing E. coli strains, which included four new ones. The CTX-M-15 β-lactamase was detected in one E. coli ST617-ST10 Cplx-B1 strain that also carried ibeA gene. One CMY-2-positive strain of lineage ST224-B2 was detected and it carried the qnrA1 gene.

Originality/value

In this study, a ST131-based virotyping scheme for strains from food of animal origin was established since this kind of strains constitutes an important vehicle of virulent ESBL- and CMY-2-producing E. coli isolates, which could be transmitted to humans by direct contact or through the food chain.

Abstract

Escherichia coli0157 is responsible for an emerging enteric disease which became a growing public health concern in 1996 with major outbreaks in Japan and Scotland. Unfortunately, the more common haemorrhagic colitis can at times develop into the haemolytic uraemic syndrome with associated renal disease which can lead to fatalities. It is now apparent that few bacterial cells are needed to spread infection and that cross‐contamination is therefore a significant problem. Although there are national programmes to investigate E. coli 0157 infection and gain new information on epidemiology, handwashing is seen as a very simple but effective approach to prevent transmission from contaminated material to food. We need to be made aware of the clinical importance of E. coli infection and how best we can bring it under control.

Abstract

Purpose

This work aims to synthesize a series of novel acyclic and/or heterocyclic systems, as precursors for dyes with potential antimicrobial activity that could be used for simultaneous dyeing and antimicrobial textile finishing. Thus, a series of novel pyridine, thiophene and pyrazolo[3,4-b]pyridine derivatives were synthesized, and their antimicrobial and textile finishing properties were studied and evaluated.

Design/methodology/approach

The synthesis, structure elucidation and antimicrobial activities of the newly synthesized compounds based on 4,4-dicyano-3-phenyl-but-3-enoic acid phenylamide (1) were demonstrated. The minimal inhibitory concentration in μg/mL of the compounds showed significant antimicrobial activity against most of the tested organisms. On the other hand, their spectral characteristics and fastness properties were measured and evaluated. Antimicrobial activities of the dyed fabrics in terms of inhibition zones (mm) were measured and evaluated.

Findings

A series of novel heterocyclic compounds (Schemes 1-3) were synthesized based on starting material (1). Compounds (1), 2, 4a, 8a and 9c exhibited comparable or even higher antibacterial activities than the selected standards (ampicillin), while compounds 2, 3c, 3d, 4a and 8b revealed higher antifungal activities than the selected standard (cycloheximide). On the other hand, some dyes showed high antimicrobial evaluation on the dyed fabrics (nylon 66, acetate and polyester) expressed as size (mm) of inhibition zones (Tables I-IV).

Practical implications

Results revealed that many hydrazo and azo derivatives were synthesized from some pyridines and thiophenes. The antimicrobial evaluation and textile finishing of the newly synthesized products revealed significant and potent values of antimicrobial activity.

Originality/value

All the synthesized compounds were novel and most of them exhibited higher antimicrobial activities than the selected standards antibiotics, thus are valuable for simultaneous dyeing and antimicrobial functional finishing of textile fabrics.

Abstract

Purpose

This study aimed to investigate how enteric pathogens and their biofilm populations on fresh produce survive according to time that contamination has occurred on leaves and contamination route: seed irrigation water.

Design/methodology/approach

Cress was contaminated in two different ways: contamination of seeds and irrigation water with 8-log MPN/mL bacterial load, Salmonella Newport, Escherichia coli O157:H7, O104:H4 or O78:H2. While contaminated seeds were cultivated for seed contamination, contaminated irrigation was applied at the end of each week to separate groups of samples obtained from cultivated surface-sterile seeds to understand how long these pathogens could survive until harvest.

Findings

The results indicated these pathogens survived until harvest, and formed biofilms on cress leaves grown using both contaminated seeds and irrigation water. No significant difference was observed among populations of Salmonella and E. coli groups in terms of survival (∼4.5–6.0 log MPN/g) and biofilm formation (∼4.4–5.7 log MPN/g) for contamination by seed. Also, SEM images revealed biofilm-like structures, the proofs of the attachment of these pathogens on leaf surfaces.

Originality/value

From our knowledge this is the first study focusing on the survival and biofilm formation of one Salmonella serotype (Newport) and three E. coli serotypes (O157:H7, O104:H4, and O78:H2), representing enterohemorrhagic and enteroaggregative E. coli pathogenic subgroups, under the same irrigation and growth schemes. Furthermore, this study mimics the contamination of seeds and irrigation water with sewage or wastewater and may shed light on contamination of fresh produce grown using poor wastewater treatment.

Verocytotoxin‐producing Escherichia coli 0157:H7 is known to cause bloody diarrhoea (haemorrhagic colitis) and, in vulnerable groups, haemorrhagic ureamic syndrome. The majority of outbreaks have been associated with the consumption of ground beef products, and cattle have been identified as an important environmental reservoir of the organism. Investigates the survival of the organism in ground beef products during normal cooking procedures. Finds that in certain products Escherichia coli 0157:H7 is able to remain viable and, therefore, maintain the potential for human pathogenicity.
